Pseudomonas aeruginosa is an opportunistic pathogen of great clinical impact. Its virulence is largely due to the secretion systems possessed by the bacteria, among which the Type 6 Secretion System (T6SS) of P. aeruginosa stands out. This syringe like nanomachine injects effector proteins into competitor bacteria to subvert their key biological functions. Here, we describe the toxic mechanism of P.aeruginosa Tse5 effector, using a biophysical approach. In particular, we show the capacity of Tse5 to form ion-selective pores in bacterial bilayers, which results in membrane depolarisation and bacterial death.
