Aluminum hydroxide (AH) has long been employed as a vaccine adjuvant although its precise mechanism of action remains unclear. In this study, we investigated the transcriptomic responses in sheep spleen following repetitive vaccination with AH adjuvanted vaccines and AH alone. After applying different treatments to sheep, among the differentially expressed genes in the Vaccine group compared to both Control and Aluminum groups, many were associated with endoplasmic reticulum stress, immune response and cell cycle. AH-alone treatment had little impact on the spleen transcriptome. These findings shed light on the molecular mechanisms of AH based adjuvants and emphasize the significance of antigenic components.
