Glutamate-induced excitotoxicity leads to oligodendrocyte death and contributes to the progression of demyelinating diseases, such as multiple sclerosis. In this study, we investigated the role of the apoptotic elements caspase-8 and its downstream target Bid in AMPA receptor mediated excitotoxicity in rat cortical oligodendrocytes. Our results demonstrate that AMPA receptor activation triggers caspase-8 activation, leading to Bid truncation and oligodendrocyte death. These findings identify Bid as a promising target to develop strategies to prevent oligodendrocyte loss in demyelinating disorders.
