Abstract

Although more than 20 years have passed since the concept of lipid raft was proposed, the existence of these nanostructures is still under discussion, as there are no noninvasive methods that can be used in living cells to investigate the nanoorganization of these structures. In this article, a bioorthogonal cholesterol probe (chol-N3) has been developed which, combined with live-cell super-resolution microscopy, is a powerful method for direct observation and characterization of <50 nm lipid rafts in the plasma membrane of living cells. This probe mimics endogenous cholesterol in synthetic and cellular membranes without perturbation.
